Clarification: If known Gram-positive organisms (like Staphylococci) appear pink, the most common culprit is over-decolorization. However, it could also be that the Crystal Violet has deteriorated or the Safranin is too concentrated. Troubleshooting Step: You must check the decolorizer (ensure it isn't pure acetone if you usually use an alcohol-acetone mix) and repeat the stain with fresh reagents and a known QC slide (e.g., S. aureus and E. coli on the same slide).

Clarification: E. coli ATCC 25922 is the standard sensitive QC strain. It must show a zone. No zone indicates a major failure.
